Framework for Kriging-based iterative experimental analysis and design: Optimization of secretory protein production in Corynebacterium glutamicum
Merge of three different aspects: (1) Current technology of microbioreactor systems, (2) lab automation, and (3) Kriging-based experimental analysis and design. The goal: maximization of the product titer of secreted GFP, synthesized by C. glutamicum, through variation of media component concentrations.

Quasi-continuous parallel online scattered light, fluorescence and dissolved oxygen tension measurement combined with monitoring of the oxygen transfer rate in each well of a shaken microtiter plate
Online monitoring of Escherichia coli and Hansenula polymorpha cultures: the device simplifies the experimental efforts, instead of parallel cultures in a shake flask and MTP, just one single experiment in MTP needs to be conducted to measure the OTR, dissolved oxygen tension (DOT), scattered light and fluorescence.

High-throughput strategies for the discovery and engineering of enzymes for biocatalysis
Novel enzyme discoveries for biocatalysis and biotransformations. Microfluidics/microplates, parallel miniscale bioreactors and others enable high-throughput screening methods and experimental design tools for screening and optimization, for the discovery, development and implementation of enzymes and whole cells in (bio)processes.

High-throughput fermentation screening for the yeast Yarrowia lipolytica with real-time monitoring of biomass and lipid production
Small cultivation systems of the model yeast Yarrowia lipolytica have been developed that mimic the conditions in benchtop bioreactors. In an microbioreactor system, a BioLector, high-throughput fermentation procedures were set up that optimize growth and lipid accumulation in Y. lipolytica.

An improvement of surfactin production by B. subtilis BBG131 using design of experiments in microbioreactors and continuous process in bubbleless membrane bioreactor
Culture medium elements were analysed by a screening DoE to identify their influence in surfactin specific production by a surfactin constitutive overproducing Bacillus subtilis strain.

Comprehensive analysis and cross-species comparison of synthetic promoters
Escherichia coli and Pseudomonas taiwanensis VLB120 were chosen as test case for the development of promoters for standardized gene expression. σ70 promoter libraries expressing msfGFP were created and chromosomally integrated into thesingle attTn7 site in both organisms.
